Could you recommend me a protocol describing TEER measurement in 2D ALI culture. I read that cell culture cup chambers for TEER measurement are used sometimes for this, but after reading the instructions I can still see that the cells must be flooded with medium / PBS from the apical side for the time of measurment. Should it be this way? After measuring just discard the medium?

Hi Marcelina,
Yes, that is the way to go. If you can borrow a fancy disk electrode, you can get really accurate measurements from the center of the culture well. It also lets you use variable amounts of fluid on the top of the monolayer.
