Hello Shazia! Dear, firstly, it depends on how much concentration you want to add according to protocol you are following. Secondly, mostly following protocols require 3 ng/ul concentration. If your requirement is same, you can prepare it by making a standard stock solution with known concentration and then pick accordingly to make your requirement.

you can combine the DAPI stain with your last antibody- solution or in the last rinsing step. The amount of DAPI-dye depends on the dilution you should use. I use a DAPI dye in a ditlution between 1:3000 and 1:5000 (0,6 and 0,4 µl/2000µl) solution. You can measure this small amount with a 0,5-10µl Vario-Eppendorf (or other) pipet. If you use a little bit more, it is ok. Much easier is it to use a mounting medium which contains DAPI, like Vectorshield (Vector Labs.)
