If a plant extract (1mg/mL) showed 28 % cytotoxic inhibition against normal cell line, then what would be its IC50.
Secondly, how can we calculate one exact dose of the same plant extract in in vivo testing, on the base on invitro test. plz guide and help me in calculations.
To calculate the IC50 (or EC50) value of plant extract in cell line, you should test several (5-6) different concentrations of it. Then you could calculate EC50 by using Hill formula.
It is not easy to calculate exact dose of something for in vivo experiments after only experiments in vitro are done. Nobody knows exactly how tested material is stable in vivo, is it bioavailable or not, how it distributes in the body, etc. At first you could test the acute and chronic toxicity in mice and then adjust a dose according to your experimental results.

if plant extract is inactive against 3T3 cell line. Will it be toxic inside animal model at same concentration.
how can we perform acute and chronic toxicity of an extract? what will be the concentration of the plant extract for this test.
