I have to isolate Tannins from methanol extract by using Sephadex LH 20 (because I have only LH20 in my lab). Can anybody suggest some protcol? I found one method but that is using 45*180 mm column, I cannot afford such a big column.
The same method will work for a smaller column. You just need to scale the volumes down in proportion to the column volumes. Assume the 45x180 mm column uses a liter of solvent. Assume you only have LH20 to make a 20x90 mm column. The volume of each column is pi*r^2*h. The ratio is
(10^2*90)/(22.5^2*180) = 0.0988 (pi cancels out). This means your small column needs ~99 mL for the same elution (1000 mL * 0.0988) with the same solvent.
