After extracting proteins out of PEGDA with Ripa and sonication i had quite good protein concentrations, but there were no bands in my Western Blot. Ponceau and Coomassie showed that there was only a undefined band around 70 kda.

1) Perform one control setup to ensure that the WB is working well.
2) 5 to 50 ug of protein concentration is good to go with.
3) If the concentration is less (very diluted sample), TCA precipitation is very helpful before WB.
