I want to isolate mitochondria from blood for further LCMS analysis
You can isolate mitochondria from activated blood cells, like LPS-activated monocytes/neutrophils.

Isolate the cells first. Mitochondrial must be kept intact for isolation.
After blood cell isolation, mitochondria can be isolated easily by differential centrifugation.
but a gradient centrifugation gives cleaner mitochondria.
