how do you isolate human myeloid derived suppressor cells (MDSC) from PBMC by MACS? Kits are available for murine cells but for human cells only FACS seems to be completely established and widely used. I’d be very grateful for any advice or literature recommendations.

one option to purify human neutrophils could be using an histopaque gradient. then you can separate PBMC (upper ring/layer) from neutrophils (bottom ring/layer). from the neutrophils ring you can further purify with MACS using CD15microbeads. MDSC are immature and stratify within PBMC ring, so you can purify from PBMC CD15+ cells with same microbeads and have a complete measure.
FACS for CD15+ purification is of course faster.

Do you want to isolate all MDSCs or each subset? I always isolate G-MDSC and M-MDSC with AutoMACS. When I want to perform an experiment with all of them, I mix these subsets in a 1:1 ratio.
