There are papers that suggest that gDNA can be isolated from paxgene blood RNA tube. I am using Paxgene RNA extraction kit for extracting RNA. We are interested in isolating DNA also for some experiments.
Can anyone help me with a standard protocol that can be used to extract DNA without the use of any Kit.
The main challenge that we face is in lysing the cells that are preserved by the preservative in the Paxgene tube.

you have to solve the DNA from the tubes. it depends on the way the nucleic acids bind on the column: Does it bind with high salt concentration, than you have to solve with low salt concentration. Does it bind with low salt concentration, you have to solve with high salt concentration.
Important is a good wash step, because contamination of RNA

I donot intend to use any kits for DNA isolation, rather would prefer to use some traditional method involving Trizol modified to suit DNA extraction from PAXgene Blood RNA tubes - Which makes it tricky for me.
But did i got it right, that you made RNA-Extraction with tubes and the DNA is still in the column?
