What are some stresses or cell death inducers used to induce cell death (apoptotic or necrotic) in human iPSC-CMs? People have used H2O2 as an oxidative stress inducer. Are there any other?
I am using human iPSC-CMs, but anything that applies to other species as well as ESC-CMs, I could use.
In my opinion, you can induce stress by exposing iPSC-CM to nutrient-depleted culture medium (RPMI), or leaving these cells in RPMI-B27 for 2 weeks. By using these approaches, you can observe cell death during prolong culture.
If you want to investigate the cell death inducers, I would suggest to use several drugs that have been well-known to target cardiomyocyte such as Doxorubicin, Carvedilol, etc. Or using high-dose of Isoprenaline or physiological dose of Isoprenaline for 48 hours to mimic heart failure conditions.

Hello Dina,
One thing which is very less tried by people and also have novelty is deprive your cells of FBS for longer durations. I have observed that as long as we increase FBS deprivation beyond 48-72 hrs, cell death get increased.
But for getting immediate cell death, people also use 10% ethanol, Co2 stress etc.
