I am facing some difficulties with the baculidirect expression kit. I use SF-9 cells with Grace insect media supplemented with 10%FBS. Cells are in good conditions. The problem is that even i do not get any CPE (tried 72-120 hours) with a control plasmid. For the gene of interest i use PCR and brightness is getting smaller (and again - no CPE).

I used to work with Bac-to-Bac and never had any problems (CPE usually occurred on P1).

LR clonase and other reagents are new, and i always follow the Invitrogen protocol.

Any suggestions?
