I have performed TLC of crude and its fraction. I got colored spot on it. I want these spot remain permanent??? what reagents should i spray or use??? also mention the how can i prepare these regents????
The liquid paraffin is a cute idea....paraffin dissolved in hexanes or dichloromethane. You reminded me that we sometimes kept some TLC plates that we had used, but we scrapped off the silica. This gave us a glass plate the same size as our TLC plate, and we used a "bulldog" clip (heavy-duty paperclip useful for many pages of paper) to hold the glass plate to the TLC plate. The glass also protected the spots....for awhile, they would still fade.
Yes Jack- it would protect but not for very long period.
Naturally the technique would be useful for hydrophilics and not lipophilics. If it is after sulfuric acid spray, we could use for all compounds . Thank you Jack for additional information. The pleasure of chromatography has to be experienced!
There are quite a number of stains available for TLC. Your compound of interest gives you the clue on the type of stain to use. Also, Uv lamp will be helpful for spot identification combined with staining technique. But you have to protect yourself from the uv rays.
