I use 293T to package the virus. A good 293T has a great influence on the virus titer. But now the 293T cells I got are unstable and they are not very good. I once obtained a very good 293T cell. Unfortunately, I did not save it well. Do you have any methods or channels to obtain good cells?
Yes, 293T cells are good for virus production, but their maintenance is key. Keeping them happy involves ensuring that they are given sufficient media (including 10% FBS) regularly and that they do not overgrow. Overgrowth will cause them to produce virus less efficiently. Try keeping them sub-confluent with the maximum density being ~85%. This would require passaging them every other day at a ~1:5. Do not let them grow in clumps either (typically due to over dilution upon splitting and they tend to form almost colony like growths).
Order a new vial from ATCC and always keep at least 2-3 good frozen vials away at -80 (for storage less than a few months) or LN2. Hope this helps...
Reuben Samson I'm not sure what you mean by over-dilution during splitting. Does it mean that the generation density is too low?
