I'm new to Western densitometric analysis, but I have watched various videos and read articles on how to conduct the analysis. My biggest problem is obtaining the peak percentage output on ImageJ. I have the box clicked when going to Analyze > Gels > Gel Analyzer Options...> "Label with percentages", yet it still only gives me area under the curve as the output on my blots.
Is there a step I'm missing somewhere?

The area under the density curve of a peak (integral) is proportional to concentration (but recall that WB is a semi-quantitative technique, see for example doi:10.1016/j.ab.2006.11.008).
You also have to select "Label Peaks" on Analyze > Gels, otherwise it only writes the AUC to the Results table even if you selected "Label with Percentages" in Analyze > Gels > Gel Analyzer Options.
