Can anyone please suggest a method for evaluating the in vitro cytotoxicity of nanometals in MTT assay. I’m trying to assess the cytotoxicity of nanometals (such as silver) but as the nanoparticles are in suspension form, and are not soluble in culture medium, I was looking for a modified method.

This is indeed an interesting question! I am also interested in assessing nanoparticules toxicity in the future and I would like to add a reflexion: how are these particles expected to interact with cells?
In order to be realistic, the exposure should be similar to the actual way in which cells and nanoparticules could meet, right? Will they be suspended in a solution as described in the paper suggested by Christoph? or will they be just dispersed in a paste? perhaps attached to a solid surface?
Certainly homogeneity has always been pursued in experimental assays and should be good to start with, but is it realistic? Couldn't the toxicity be related to anisotropy or insolubility?
I agree with Dr. Mullick Chowdhury. Also, trypan blue, although tedious, is very reliable. It will give you a better assessment of viable cells compared to most enzymatic assays.

for your MTT analysis you can also try making nanoparticle solution in DPBS and incubate them with cells for your optimised incubation time, and then narrow down your results with other cytotoxicity assays like LDH assay,,,,,,
