I want to graft GFP+ stem cells by intravenous injection into mice and follow their distribution in various organs (blood, brain, liver, lungs, spleen, bone marrow...), at different time points. My lab is not equipped for radioactive work nor bioluminescence and immunostaining would be too time-consuming given the large number of organs. I was thinking of using qPCR to detect the gfp gene, or Western blot against GFP (but is it quantitative enough?), or even flow cytometry after dissociating the different organs. Any advice? Does anyone have a good protocol to recommend?
