Can anyone please tell me how one would add the amount of dmso necessary for my vehicle controls for cell culture drug treatments? For example, i have four drug concs: 0nm, 200nm, 500nm, 1um. I believe I treat the 0nm group with 1um of dmso, and the 1um group with 1um of the drug. If i treat the 500nm group with 500nm of the drug, do I have to also treat that group with 500nm DMSO, how would I do so?

You will have to use matched DMSO concentration vehicle controls.
You will be preparing your drug in 100% DMSO (stock solution). You will then make further dilutions from the stock solution using cell culture media or sterile PBS to obtain the working concentrations namely, 1uM, 500nM and 200nM.
So, for 1uM, for instance, you will be diluting your stock say 100 times with culture media / PBS. At the same time the DMSO will also get diluted 100 times for this concentration. Thus, the DMSO concentration as vehicle control for 1uM will be 1% DMSO because DMSO has been diluted 100 times.
So, you will dilute 100% DMSO 100 times using culture media to obtain 1% DMSO and add equal volume of this 1%DMSO only to the vehicle control well for the (1uM drug + 1%DMSO). In this way, the vehicle control well will match the test well in terms of DMSO (1%).
In a similar manner, you may proceed with the other two concentrations, 500nM and 200nM. For these two concentrations the DMSO vehicle control will be further diluted. So accordingly you may do the needful.
For 0nM group, however, there is no drug added. The well will consist only of cells in culture media.
You need to bear in mind that the final DMSO concentration in cell culture should be 0.1% which is considered to be safe for almost all cells. So accordingly prepare the stock solution so that when you prepare working solutions, the DMSO concentration is equal or less than 0.1%.
