I agreed with Premont explanation as the calculation of collagen concentration is crucial due to easily degraded composition. To best of my knowledge and experience you could use the Sircol Assay Kit to quantify the collagen composition as this kit is really helpful and specific protocol for you to quantify from different sources including tissue, cells and others.
