We want to test the effects of PTU on function of cells. Although literature data decribe that PTU can be dissolved at a final concentration of 5 mM in cell culture medium, our first trials show that it does not dissolve. A soultion in DMSO is possible, but after adding to the medium precipitation occurs.
Does anyone has a procedure for this?

Based on this data, the best solution to enhance the drug's solubility is by heating the medium's buffer or increasing slightly the pH.
Propylthiouracil is a weak acid which can be ionized in basic medium. Increasing the pH from 7.0 to 8.0 is expected to significantly enhance the drug's solubility.
Alternatively, you can dissolve it in hot water, then add the buffer components and adjust the pH (or dissolve it in the buffer with the aid of heating)..
