I wanted to get 0.2 mg/ml Ferrostatin-1 and 0.1% DMSO respectively. 10mg Ferrostatin-1 was dissolved in 50 µl DMSO first（in fact it was undissolved）, and diluted with 50ml 0.9% NaCl. Then the tube was warmed at 40 ℃ and shaked in the ultrasonic bath for one hour.After that I found Ferrostatin-1 was still undissolved. So would you give me an advice to dissolve it?

Try to dissolve ferrostatin-1 in DMF solvent first and then dilute it further with buffer such as PBS.

According to the Ferrostatin-1 factsheet (avaiable at https://www.sigmaaldrich.com), it seems that you may have failed to dissolve ferrostatin-1 because the concentration of your solution is above its solubility threshold (which is 10 mg/ml of DMSO according to the manufacturer). Maybe could you consider dissolving it in 1 ml DMSO instead of 50 µL, if this is still suitable with your experiment. You may have higher amount of DMSO than desired, but you can balance this by adding the same amount of DMSO to your control solution.

Thank you very much. I have rescued the depressed experiment. As Marius Baguma kindly advice, it's impossible to balance the minimized DMSO concentration (0.1% ) and maximized Ferrostatin-1 final concentration (0.2mg/ml). So the turbid liquid was centrifuged and the sediment was extracted. I used 0.5 ml DMSO to dissolve the sediment, and it worked. I added the solution to the liquid supernatant. The final concentration of Ferrostatin-1 and DMSO were 0.2ml/ml and 1% respectively.
