I have isolated plasmids from bacterial cultures, now they are of no use to me. How should i discard them?
Recombinant nucleic acid-containing materials (i.e. cultures, microbes, plasmids, animals, plants) must be decontaminated or inactivated before disposal. Chemical treatment with an appropriate disinfectant is an acceptable method of inactivation. Autoclaving of rNA-containing materials is also acceptable.
Send the sample to us if you have no use for plasmid isolated. we train students they will get help.
I think the required procedure depends on the nature of the genetic information on the plasmids.
I believe you must consult with the safety adviser at your institute.
Autoclave the plasmid DNA,
If your plasmid is labelled with isotope, discard it using a radioactive-material-dispose standard protocol.
Attached is the guideline for Safe Operating Procedures for bio-waste in the research universities of the US.
Use 0.25% Sodium Hypochloride to inactivate and Autoclave it to discard it.
Though most of the bacteria are unable to survive in the absence of suitable media yet one may treat them with lysol, bleach, or alcohol before discarding them, as it will be safer to do so!
