I wanted to know about detection tests beside API or molecular techniques which can differentiate lactobacilli species

Please, you can use biochemical methods for identification of lactobacilli. Good ref. Bergey’s Manual of Systematic Bacteriology (2009), Volume Three, Page 465
Hi....you could try sugar fermentation test for different species using bergeys mannual. or species specific PCR amplification of16s rRNA gene as this region is highly conserved.

usually we use 3 simple test to identify Lactobacillus
1) typical growth on MRS agar
2) negative growth on the microbiological agar
3) catalase negative reaction on the slide
HI you can go for the typical biochemical tests for the Lactobacillus sp. as mentioned in Bergey's Manual
