Surely, tumor cells from the tumor site in patients are lack of normal function of p53, the immortalization is essential.

I do not understand why a cancer cell line derived / established from a patient needs to be transfected for p53.
It seems to me that this process can modify the behavior of the cell line.

Why are "new cell lines" needed according to the fact that hundreds of them are already well characterized (see the attached article).
