I am working on the assessment of the efficacy of several new vitamin D analogus in treating postmenopausal osteoporosis in rat model. Postmenopausal osteoporosis was induced by performing ovariectomy (OVX) in 3 months old female rats. By literature review, there have been two kinds of setting for initiation of drug administration: 1. Drugs are used just after OVX operation or a week later for wound healing. 2. After OVX operation, the rats are kept for 12 weeks to induced obvious osteoporosis. Then, drug administration begins. Most literatures took the first setting, setting 2 is also adopted by some studies. It seems that setting 1 is focusing on the protective effect of drugs on bone mass whereas setting 2 is mainly on the ability of drugs in correcting already established osteoporosis. I am wondering which one is more rational and suitable for my experiment design.
