A part from OD measurement and FCM methods, do you know others trustable way to determine the biomass growth in media that have particles in suspension and precipitation and/or very dark colour?
Several methods requiring destructive testing are available.
1. Monitor the change in dry solids and calibrate dry solids with biomass content.
2. Measure spectroscopic signature and monitor relative peak change in emission between C-H regions and regions related to the dark material and suspended particles. It is quite possible that the dark material will be transparent in some regions of the spectrum and you may be able to do this in transmission instead.
3. See if electrical conductivity, speed of sound, density, or viscosity correlate with biomass content and measure these.
Collect all the biomass by centrifugation in a pre-weighed container and use a digital balance to weigh the container + biomass. Subtract the weight of the empty container to get the biomass. If the biomass can be collected by filtration, use the same idea with a pre-weighed filter.
