I have performed serial dilutions of my sample to get different concentrations and inoculated the bacteria using a 96 micro well plate. I took the OD600 using multimode reader. Now, Shall i determine MIC and MIC-50 from the readings i got or shall i take MIC as visual observation ?
Hi Ubaid,
It is always preferable to use quantitative methods to accurately describe your results, so I would definitely use the OD readings already taken to calculate the IC90 and IC50.
Doing so will enable you to more accurately compare any future results (for example if you test against a variety of other bacteria) as the data will be consistently corrected against "normal" baseline growth for the particular organism in the conditions you use.
Visual MIC determination is great for quick applications and clinical labs, but if you have access to a plate reader use it to its fullest!
