I'm wondering if there are "classical" bacterial outer membrane vesicles markers similar to tetraspanins (CD9/CD63/CD81) used for human/animal cell exosomes that could be detected by WB or ELISA or similar.
OmpA and Lpp seem viable options, but might bias the bacterial species whose OMVs are detected. Peptidoglycan or LPS are more "general", but obviously their detection is not necessary OMV-associated.
Is there something better?
As far as I've read, there's no universal set of proteins associated with OMVs in bacteria. I think they're usually isolated using density centrifugation/molecular weight cutoff filters and detected with lipid staining dyes.
Thanks for your answer! I was hoping there's a way to differentiate them from other lipid particles in complex sources at least in bulk, but looks like there's no easy way for this.
Hi. You could do a SEM of your exctracted OMVs to be sure that your sample correspond only to OMVs and not a mix with bacteria o debris. And, run a SDS PAGE first to see if there´s a region with more bands to use it to follow your extraction or find a marker.
