I would also recommend using existing systems that have been well-tested and published. I use the Applied Biosystems TaqMan system and sometimes Exiqon. However, some feel they can save money by designing and ordering their own primers. I agree with Paulo, however sometimes you need to use your imagination for example with miRNAs which are not already well characterized. In this case, try to apply the protocol described in this paper. Hope it helps
