Hi everyone!

I’m wondering if you could give me some advices regarding my data analysis. This is a new type of analysis for me…

I’m using Imaris for quantification, I have 5 different channels in 1 photo. Some of them are DAPI staining (all cells) and Ki67 staining (negative - G0 cells, positive - non-G0) and CD38 (HSCs) What I need to do is to understand if my HSCs are G0 or non-G0. So they have to be CD38+ DAPI+ Ki67-/+ The problems are:

  • CD38 is surface marker and DAPI and Ki67 nuclear. So if it’s possible to define G0/non-G0 for my cells at all? Most of them are Ki67- so maybe it’s easier just to look with my eyes?
  • If cells are non-Go (so they are Ki67-) how can I detect it? I can’t create Spots with negative cells? I think the perfect decision is to create Spots based on DAPI+ Ki67- and Spots DAPI+ Ki67+ but is it possible at all?
  • I hope I didn’t confuse you, but I’m here to give more details
