I have problem detecting specific proteins using Western Blot. I am pretty sure that it is not the problem of my technique or antibody since I can still see loading proteins and ladders on the blot.
I am assuming that protein expression of interest is too low to be detected on the blot in the cell line I use. The protein is tumor suppressor and I was able to see the upregulation of this RNA after histone deacetylase inbhitor treatment.
When I run PCR these RNA start to show up around 27-28 amplification cycles. I tried to load lysates from 20-40 ug but I was still not able to detect the protein of interest using western blot.
Have you experienced this problem? What is the standard amplification cycle of RNA in qPCR to be detected on Western Blot?
If I cannot detect on Western Blot, is there any alternative to detect or support the low expression of these proteins?
