I have cultured thermophiles bacteria via the pour plate method. As of now, I am trying to culture pure strains of bacterial colonies, It is not able to grow. Kindly please suggest something??

If you are streaking for isolation, you could try limiting dilution with pour plates as they grew with this pour plates initially.

Anjali Patil .. Additional information would be helpful to give good suggestions. If you have isolated a bacterial species (strain) but it is not growing in pure culture the first thought I have is that it requires some metabolite from another bacterial species that co-occurred with it in the hot springs. There are several ways to see if this is the case.
Anjali Patil .-. Have you tried either of our suggestions and did you get acceptable results? It would be nice to know what if anything worked.
