I am using Ficoll Plaque based isolation of mesenchymal stem cells from umbilical cord blood. I see reduced cell numbers after Day 6. What would be the key factors to account the quality of cells? What is the preferred media being used to culture MSCs and how long does it takes to haves MSCs?

The number of colony-forming units in umbilical cord blood is relatively low and reduces further in the full term of pregnancy (https://www.nature.com/articles/pr2008147). This is the reason for reduced success rates of isolation.

)You can use DMEM:F12 containing 10% FBS, 1x Glutamax or use MesenCult™ Mesenchymal Stromal Cell Culture from Stem Cell Technologies for better results.
Make sure the cells are not sparsely populated during isolation. Use smaller dishes (60mm or 35mm) if the yield is low. Later it can be expanded to larger dishes (100mm).
