I have two images, taken in the same camera, one taken with a 10X objective and another with a 5X objective. The pixel/micron ratio for the 10X image is 1px=1.25um. But, I do not have a calibration image for the 5X objective. Can I get the pixel/micron ratio for the 5X image from this?

If you are using microscopy, the scal specification will be available either in soft copy or in the manual. Read it carefully. If you can mention what instrument, you are using, it will helpful to calrify your doubt.

It should be proportional, according to the definition of magnification.
