Hi, I would like to try to express a fusion protein of LAMP-1 and a fluorescent protein to direct the fluorescence to lysosomes. The fluorescent protein will be fused with the C-terminus of LAMP1, so that it finally remains in the cytoplasm.

LAMP-1 has a pretty big lumenal domain. I was wondering whether I could exclude it from the final construct. It will make the fusion gene 1kb shorter and ease the subsequent overexpression in primary cells.

I was wondering, can design the construct as simple as "LAMP leader peptide+ LAMP transmembrane domain+ LAMP cytoplasmic domain + spacer + fluorescent protein", or should I add few amino acids of the lumenal domain for proper folding? Like "LAMP leader peptide + last 3-4 aa of the LAMP lumenal domain + LAMP transmembrane domain etc." Thanks a lot!
