Agrobacterium mediated Virus infection in host plant resulted in visible Hypersensitive response at 2nd day after virus inoculation. Can any one suggest me the optimal time intervals to study the ion leakage?
How to take the readings?
Do I need to use the same sample at different time periods or different one for measuring the conductance?
A general comment on experimental design. When approaching a new system take your first set of samples at exponential intervals or dilutions you can cover the most range efficiently. In the future you can zoom into zones that look intersting. Start by asking how many samples can you reasonably handle? Then ask what the minimum and maximum times are. Finally ask which kind of exponent to use, e.g. 2 fold, ten fold, or some other.
