I am working with a cell line (EA.hy296, HUVEC hybrid) for the first time. How do I know, which passage is right to work with? I dont find any passages in publications except one thata says use passage 3-6, but that seems low for a cell line? Or doesn't it matter because it's a cell line?

there isn't a generalized rule to work with cell lines, each specific type may have its own optimized work conditions. Nevertheless I do recommend the following:
1) After thawing cells, passage them at least three times, for them to recuperate their morphology and specific expression panorama. If you take a look at the first few passages, the cells may not have the morphology you expect for them.
2) Do no you cells in passages above p20, as they may have been more susceptible to mutation.
