I have a related research which is refers to fabrication of nanofiber tubular structure for artificial blood vessels. Since I have to increase antithrombotic characteristic in my tubular structure, I decided to add some anticoagulant dissolution to enhance my tubular structure antithrombotic ability, but I do not know its cytotoxicity existed or not, my professor asked me to add this dissolution in media and use this media to culture cells, did someone has this idea about doing this thing and could tell me how much amount of dissolution I should add in the media?
There is a number of ways to evaluate the cytotoxicity most common and easilly accepted is MTT assay. However if you have a very limited number of cells or no compatible plate reader you can use Propidium iodine staining to detect the dead cells. I am not familiar with the tubular structures you are working with, if you know the concentration of anticoagulant you have in your system then use the same concentration in culture medium. Otherwise, you can search the literature for reports of anticoagulant-induced cell toxicity and pick up a relevant concentration range. If possible you should test several anticoagulant concentrations on your cells.
As Yanling suggested, you can use propidium iodide staining to detect dead cells in 96-well plates, given that you have access to a microscope to count cells. Otherwise, you can still perform a resazurin-based assay with a simple plate reader to assess cell viability in your wells at a more global scale.
