I have alcalase enzyme it's activity 10000U/g and I want to use it in hydrolysis whey protein isolate 90% protein. A suspension of 5% (w/v)WPI prepared so I am asking about the quantity of enzyme need to be added.
The exact answer is not obvious. You must consider your flask as a reactor. By knowing the kinetics and kinetic parameters of your enzyme, you should be able to calculate the required enzyme load (see the enclosed file). However, for proteases this may not be easy. This, because of the substrate changes during the reaction. Thus, I suggest you try with the enzyme dose suggested by the supplier.
