I am using Elabscience sandwich ELISA kit (detection range 0.31-20ng/ml), I am using supernatant saliva samples. I am using 4 plates and plotting the standard curve for every plate. In one of the plate, I have plotted the standard curve and according to the linear part of the standard curve, the cut off OD is 1.8. I have samples which have OD greater than 1.8 (max OD of one of the sample came out at 2.6 (conc 36ng/ml). I know that I have to dilute the samples so that the OD comes within range. Samples that need to be repeated have conc between 8 - 36 ng/ml (OD 1.82 - 2.6). How do I determine the dilution of the samples based on this information? I was thinking 1:3 (25 ul saliva + 75 ul diluent) = dilution factor 4, will this be fine ?
