As a part of my process, I need to freeze my protein solution in 96-well plate and then freeze dry it. I need to remove the solution intact to study it further. But the scaffolds stick to the wall and I am not able to take it out. Any suggestion?
The sticking of the scaffold occur due to the cell culture plate polymer and the coating treatment . the cell culture are normally polystyrene which are treated/coated (e.g with collagen, poly-D-lysin) for better adhesion of cells. polystyrene is a hydrophobic polymer but the coating make it hydrophilic. I think you can try the untreated 96 well plates which are also available. the untreated plates are more hydrophobic which probably prevent sticking of the sample after freeze drying.
