I'm trying to assess pAMPK/AMPK using western blot, which has been published in lots of literature. But I find the MW of both pAMPK and AMPK are 62KD. I think it is impossible to stain these two proteins on the same membrane. Also I didn't think I could analyze the blots of pAMPK and AMPK on two different membranes to get the ratio, unless pAMPK/beta-actin and AMPK/beta-actin are presented to declare that the blots of pAMPK and AMPK are from the same amount of proteins. But I haven't read about this part in any literature yet. Maybe I have made some misunderstanding. So what shall I do? Can anyone give me some suggestion? Thank you!
