However more information are needed to better answer your question. I would say that is pretty hard to have equal virus inoculation, but you could use PCR methods to know how much virus content you have in your total DNA.
when you make inoculation, as you know you prepare virus inoculum with grinding different or another plant tissue. After extraction, the sap inoculum includes definite amount of virus if extracted plant is infected with wanted virus.
Obtained extraction includes same virus concentration. When you take definite amount from this extraction (ie: 0,5, 1ml or 2ml) and use it for inoculation, it means that same amount virus is inoculated.
The most important thing in your question is time intervals. Virus can lose its stability in extraction in time. So fresh extraction had better to be used permenantly. I hope my explanation is clear for you.
