I am using rink amide to synthesis a peptide with c-terminal as CONHCH2CH3. After cleavage the peptide from resin I will have CONH2. How can I change it to CONHEt with this regards that I have Arginine, serine and histidine in the peptide sequence.
If I react Rink amide before synthesis and load a Ethyl group on resin, will destroy the resin in reactivity? and will it dissolve my problem?
use 2-chlorotrityl resin and after the cleavage do the ethyl amidation with ethyl amounium chloride and a base like NMM.
first of all, how would you attach en ethyl to the rink amide ?....reductive amidation of a 3 amino propanal?....anyway then it will not cleave with TFA....I suggest you to use a linker suitable for the synthesis of peptide thioesters, and perform aminolisis with ethylamine post synthesis...
