I am trying to generate a pure population of lentivirally transduced primary human CD4 and CD8 T cells.

My main obstacle at this point is the last purification step. I have already successfully infected and expanded cells that express my genes of interest. The main problem is, after selecting for puromycin resistance (conferred by lentiviral vector), my T cell culture contains a LOT of dead cells.

Centrifugation techniques have not been sufficient in my experience, as many dead cells are still spinning down with my live cell pellets even if I do 200g spins for 3 mins. This method has also been more prone to live cell loss.

We have a flow cytometer, but not a FACS cell sorter. Has anyone tried negative selection kits to remove dead cells for this purpose?
