The upside of using spores is that you can inoculate a known concentration by counting the spores in your spore suspension (either by using cell counter or diluted plating and counting CFUs). This way you can compare your wells and treatments fairly

I guess you could also use vaccuum filtration on a liquid culture, catch the mycelium on a membrane, measure dry weight, and standardize using the dry weight.
