Our lab develops antifungal drugs. I'm developing an assay for detecting the effect of antifungal compounds on the growth of Colletotrichum dematium. My plan is to grow liquid cultures of C. dematium in a 96 well plate along with an antifungal drug. Then I'll add resazurin and determine growth by measuring fluorescence.
My question is: how do I prepare the C. dematium culture - should I make a spore suspension? Or could I just grind up some mycelium growing on a petri plate then dilute it to make the inoculum? I would be easier to do that than having to collect spores.
