I want to dissolve my Ibrutinib in pure DMSO, it can be stored at -20°C according to the company. They unfortunately do not specifiy if there is a limit to thaw and refreeze the drug without loosing activity. Is there a rule for drugs????

In general, when any compound is dissolved in DMSO, the solution should be frozen into small usable aliquots for a single use for all experiments (if necessary, the left over solution should be combined at the end for less important and pilot experiments).

I agree with Divaker. However, we asked ourselves the same question and found that if we kept the remaining Ibrutinib at 4oC and used it within 48h after thawing the ibrutinib retained full activity.
Always use fresh stocks to do your experiment. Every freeze thaw cycle reduces the potency of compound.
In my experience, small aliquots should be used for each experiment and not refrozen after thawing, in agreement with other responders. Also (again, in my experience), when diluting Ibrutinib out of 100% DMSO in order to use with cells, the dilution out of 100% DMSO should be made just before each experiment. Ibrutinib seems to lose potency after even a short amount of time (30 mins) when diluted out of 100% DMSO, even when kept on ice.
