I am currently trying to analyze ROS response of cultured cells (HT-29) to mice feces. I homogenize 1 mouse poop in 2 ml of DMEM, split in half and filter sterilize 1 ml and heat kill and filter sterilize the other 1 ml. Then I put 50 ul of either fraction in 200 ul on a 96-well plate, and measuring ROS response after some time. I wonder if there are more standardized protocols for tissue cell response to feces, and whether I am putting too much or too little feces.
