Are you planning to add drug in the polymer matrix. If yes then the property of the drug may affect the chitosan to TPP ratio. In that case you  may have to optimize the ratio.

Hi Sawara, probably you are using concentrated acetic acid solution (i.e., 2% acetic acid solution is probably high concentration for your chitosan). Again if you want to add drug in the solution that will affect the solubility of chitosan as well as the ratio between chitosan to TPP.

We were synthesized the chitosan nanoparticles (150-200 nm) by using low molecular weight chitosan (0.2%) in acetic acid solution (0.2%) containing P407 (0.6%) and then TPP was added dropwise in to the above solution (Cs/TPP= 4/1, pH=3.5).

what happens if I add TPP concentrations > 1 mg/mL (like 3 mg/mL) to a 2,5 mg/mL chitosan solution in acetic acid 2%? I have already obtained a milky solution, but dont know about the size of the NPs.

A lot of factors are involved in confirming the particle size. Particle size also depends upon the stirring rate and duration i.e., 3000 rpm for 10 minutes or 20 minutes.
