I want to measure drug relase of my compound in polymeric nanoparticles. But the problem is my drug cannot dissolve in pbs. Most papers use pbs as release media. Can i add 20% dmso in pbs to help my drug dissolve well? Or is there any method to solve this problem?
If your drug will not dissolve (even slightly) in PBS then it cannot be used as a treatment for any living creature. DMSO is an aggressive and very unpleasant solvent - it could never be considered as a vehicle to dissolve anything to be injected into a living body: I'm sure you'd never experiment on yourself with a shot of DMSO..... You need to find some other drug delivery mechanism - perhaps a stealth liposome where the drug may be more soluble than in PBS.

I and some other researchers experienced similar challenge in the past. What we did was to conduct the release study in a surfactant medium inorder to facilitate the release of the drug from the polymeric NPs.

You can try tween 20+ PBS. Tween is a surfactant that can help dissolve hydrophobic drug; however, high percentage of tween can also disrupt your nanoparticle structure. So you may need to vary tween concentration in your PBS solution. Tween 0.05% in PBS is commercially available.

You can add surfactant such as sodium dodecyl sulfate into the PBS medium.
