I am trying to find how many samples can you go through with Enzymatic Assay method ?
(assaying l-citrulline and l-arginine)
I am using this research article to find the information but I can't find the answer, can someone have a look at it and explain me how many samples does the author of this research paper gone through?
The assay was apparently performed in a microplate format and monitored using a plate reader. Such assays are typically performed in 96-well plates or 384-well plates. Details are not given about the volumes or the plate type or the number of samples. However, the authors mention that they had 20 blank samples. I think that they probably used a 96-well plate.
Since the assay requires continuous monitoring, the maximal number of reactions that can be run at once is equal to the number of wells in the plate. Thus, 96 reactions, including controls and blanks, could be run at once using one plate reader. The authors do not say for how long the reaction is monitored. If it is not very long, one could probably run multiple plates of reactions in a working day.
Adam has given you an insight into the assay. One can perform multiple assays depending on the availability of the 96-wells plate reader or a uv-visible spectrophotometer. The microcuvette of 100 microliters in the absence of a plate reader can be used to perform multiple assays.
