TRIZOL protocol allows extraction of total RNA content, including small RNAs (microRNAs etc). When there are no specific needs to extract the only small RNAs, TRIZOL is highly recommended, in particular for extraction from tissue specimens.
The methods that use a column like miRNeasy kit and miRVana are a good choice. Trizol is good, but we must be careful, because recently was reported that miRNAs with low GC content, are selectively lost during sample preparation rather than degraded in the cell. The small RNA loss occurs when a small number of cells is used for RNA preparation using the standard TRIzol protocol.
